Although oral delivery is a more challenging route for fragile mRNAs, its feasibility has been demonstrated in rodents and pigs using ingestible milli-injector capsules, holding great promise for the development of oral mRNA vaccines. BioNTech and Matinas BioPharma recently announced an exclusive research collaboration to develop potential oral mRNA vaccines using a novel lipid nano-crystal platform. This lipid nano-crystal—a stable crystalline nanoparticle containing multiple layers—is formed via the interaction of calcium and anionic phospholipids, during which active drug molecules inducing mRNAs can be loaded within the layers.
